Establecimiento del hongo comestible (Agaricus sp.) bajo condiciones de laboratorio para su multiplicación en diferentes medios de cultivo

Abstract

Edible mushroom production has two different stages: the vegetative stage and the fruiting stage. The vegetative phase is performed in a biotechnology laboratory and covers the technique for obtaining “spawns”, which parameters include the multiplication and reproduction of the mycelium. The fruiting phase begins with the appearance of edible mushrooms and includes everything that occurs outside the laboratory. In our country, production of edible mushrooms is limited and generally unknown. So, in this study, the vegetative phase was divided into two stages and conducted in the laboratory. Stage 1: We inoculated spores and implants of the edible mushroom species Agaricus in three synthetic growth mediums: PDA (Potato-Dextrose-Agar), PDY (Potato-Dextrose-Yeast), and MEA (Barley-Biphosphate Potassium-Agar). These were incubated in different growth chambers at three different temperatures (17ºC, 20ºC, and 25ºC). The best mushroom development in terms of micellar growth was obtained in the PDA growth medium. The temperature that contributed most favorably to this development was 17ºC. Stage 2: We re-inoculated implants from the crops of the previous step in four natural substrates (brown rice, barley creole, brown rice combined with horse manure, barley combined with horse manure) and incubated them in growth chambers at three different temperatures. It was observed that the best micellar growth occurred in the natural substrate containing barley creole. Also, the most effective incubation temperature was 20ºC. Thus, we established that the barley grains sold in our city work well as a cheap natural substrate to propagate and produce edible mushroom “seed” of the Agaricus species at a temperature of 20ºC.

La producción de hongos comestibles en general, comprende una fase vegetativa y una fase de fructificación. La fase vegetativa se realiza en un laboratorio biotecnológico y abarca toda la técnica para la obtención de "semilla de hongo", cuyos parámetros de multiplicación y reproducción de micelio es conocida también como obtención de cepa pura de hongo. La fase de fructificación inicia con la aparición de las setas u hongos comestibles y todo 10 que implica las prácticas fuera de laboratorio. En nuestro país esta producción es limitada, y por 10 general desconocida, por ello en la presente investigación, se realizó en laboratorio la parte que implica la fase vegetativa, que a su vez la dividimos en dos etapas. Etapa 1: Inoculamos esporas e implantes del hongo comestible Agaricus sp. en tres medios sintéticos de cultivo PDA (Papa-Dextrosa-Agar), PDY (Papa-Dextrosa-Levadura), MEA (Malta-Bifosfato potásico-Agar) incubados en cámaras de crecimiento a tres diferentes temperaturas (17°C, 20°C, 25 °C), y el mejor desarrollo del hongo en 10 que se refiere al crecimiento micelar fue obtenido en el medio PDA, y la temperatura que contribuyo favorablemente en este desarrollo fue el de 17°C. Etapa 2: Reinoculamos implantes del cultivo anterior en cuatro sustratos naturales (granos de arroz integral, granos de cebada criolla, granos de arroz integral combinado con estiércol de caballo y granos de cebada también combinada con estiércol de caballo) incubado en cámaras de crecimiento a tres diferentes temperaturas. El sustrato natural en el cual se observo el mejor desarrollo micelar fue en los granos de cebada criolla y la temperatura de incubación con mejor efecto fue de 20°C. De esta manera se pudo establecer que los granos de cebada que se comercializan en nuestra ciudad son buenos como sustrato natural y económico para propagar y producir "semilla" de hongo comestible Agaricus sp. a una temperatura de 20°C.
